A plea of no contest has been entered into evidence for a second time in Hennepin County District Court against a woman accused of stabbing her boyfriend multiple times.

Mykolas Varnas, 19, of Hopkins, was found dead on July 9, 2011, of stab wounds on his torso, both arms and right side of his back.

A medical examiner ruled the cause of death was multiple sharp-force injuries and ruled the manner of death a homicide.

Varnas was found in his home by his roommate. A friend knocked on the door of Varnas’ Southdale apartment, and when no one answered, the roommate then checked the back door and saw Varnas’ lifeless body wrapped in a sheet.

At a pretrial hearing, the Hennepin County attorney’s office argued that it was improbable that the sister-in-law of Varnas’ roommate, Mykola Demetras, staged the scene, and that her lying to police at the scene was indicative of her involvement.

Citing the need to protect her child, Demetras gave a false statement to police. She said she was home alone with her two children when she heard the doorbell ring three times. She then answered the door and saw Varnas, who she met for the first time that night. Varnas asked Demetras to call 911 for him.

The report of an officer who responded to the scene said Demetras told her son to hide behind the shower curtain while she told police Varnas had never called her before and that he asked her to call 911.

Varnas was stabbed multiple times, and when police arrived, he was lying with his hands wrapped around his stomach.

Before an assistant Hennepin County attorney said that the plea of no contest was entered, Judge Robert Scheibner asked a woman at the back of the courtroom if she is Varnas’ sister-in-law.

She said no.

The attorney went on to say that the plea of no contest was entered on Varnas’ behalf by the woman’s husband.

“Is it accurate, that you’re actually the sister-in-law of Mykola Varnas?” the judge asked.

“I am,” she said.

“Even though you were the one who told police that you didn’t know him?” he asked.

“Yes,” she said.

Our initial goal in this line of investigation was to search for novel pathways and cell signaling proteins involved in the production and transport of the G-CSF-receptor in AML-1 cells. We have successfully localized the G-CSFR to the plasma membrane and have found that G-CSF acts to activate and internalize the G-CSFR in a clathrin-independent fashion. As a consequence, the classic signaling pathways such as the MAPKs, the AKT and the JNK are not activated by G-CSF in AML-1 cells in contrast to other cell types, and the JAK-STAT pathway is not activated. Another novel observation was that cytochalasin B, a well-known inhibitor of actin polymerization, is a potent and specific inhibitor of G-CSF-induced JAK-STAT activation in AML-1 cells. We are continuing these studies and have isolated the G-CSFR gene from AML-1 cells and found that it is mutated at tyrosine 839. The mutation results in aberrant G-CSF signaling and altered G-CSF-induced growth and cytokine production. G-CSF plays an important role in the hematopoietic system and has been used as an effective cytokine in the treatment of various hematological disorders and solid tumors. However, despite all the therapy, mortality of cancer patients and patients suffering from hematological disorders has not been decreased significantly. Therefore, to increase the efficacy of G-CSF as a cancer therapy, we searched for an improved way of administering G-CSF with a lower dose of G-CSF and found that leukine could act as a substitute for G-CSF to upregulate the expression of G-CSFR in the surface membrane of AML-1 cells. Our new findings indicate that leukine can replace G-CSF in vivo to upregulate G-CSFR expression and to elicit potent cytokine responses. The new protocol of treating cancer patients with a combination of leukine and G-CSF is being evaluated in order to improve the efficacy of G-CSF. We have shown that AML-1 cells express the IL-6 receptor subunit gp130, and have identified the STAT3 and STAT5 tyrosine phosphorylation sites. In addition, our preliminary data indicate that G-CSF strongly activates IL-6 signaling in AML-1 cells through the gp130
