Protein-rich extracts of microorganisms, plants, and animals have traditionally been fractionated by metal affinity chromatography (IMAC), which suffers from a nonselective elution of metal-associated proteins and by size-exclusion chromatography (SEC), which is tedious and requires costly disposable columns. Here we introduce a novel method for extraction of microbial biomass and fractionation of biomass into high- and low-cadmium-affinity compartments. The method requires a lysate preparation that is mixed with a low-molecular-weight-impermeable but metal-binding polymer, which, in the presence of EDTA, selectively traps metal-associated proteins in the interstitial space between polymers. Spent lysate can be mixed with a fresh lysate for a single-step extraction, with columns (20 cm × 1 cm) requiring a total volume of 0.5-1.0 L of spent lysate. IMAC of the spent lysate results in a high recovery of iron and nickel and a 30% recovery of zinc, whereas a 1-day SEC of a batch of spent lysate yields a fraction with ∼0.3 mg of cadmium per
